Giredestrant and everolimus extended progression-free survival after CDK4/6 treatment

In the phase III evERA trial, giredestrant plus everolimus extended progression-free survival for people with advanced ER-positive/HER2-negative breast cancer whose disease had progressed after CDK4/6 inhibitor treatment, MedPage Today reported. Among patients with ESR1-mutated tumors, median progression-free survival was 10.0 months with the combination and 5.5 months with investigator-selected endocrine therapy plus everolimus.

The trial included 373 patients from 13 countries, and ESR1-mutated tumors accounted for 55% of participants. In the overall population, median progression-free survival with the combination was 8.8 months. Any-grade adverse events occurred in 97-99% of patients, and fatal adverse events occurred in 2.7% of each group.

The estimated 18-month overall survival among patients with ESR1-mutated tumors was 71.5% with giredestrant and everolimus and 50.9% in the control group.
